可以請大家幫我比較以下2個protocol嗎？
對於抽8cc有沒有什麼問題？哪個比較適合？
RBC ＆ Cell lysis buffer需不需要加量？

protocol 1:
Cell lysis step:
1. 3ml blood(EDTA) + 3 vol. RBC lysis buffer to 15ml centrifuge tube.
Invert tube, mix well, incubate 10min at RT.
2. 2500rpm, 3min., 4C
3. remove sup., resuspend well white cell pellet.
4. add 3ml cell lysis buffer, mix well at RT.
(if cell clumps were visible after mixing,
incubate 37C till solution is homogenous.)
Protein ppt step:
5. add 1ml 10M NH4OAc to cell lysate.
6. vortex, high speed, 20s. incubate on ice for 5min.
7. centrifuge 3000rpm, 15 min.(repeat 6,7 if pellet not tight)
DNA ppt step:
8. discard sup. to clean tube, add 3ml 100% isopropanol.
9. invert gently, mix well, until white threads form a visible pellet.
10. 2000rpm, 3min.
11. add 3ml 75% ethanol, invert tube(wash DNA).
12. 2000rpm, 1 min, carefully pour off ethanol.
13. air dry.
DNA hydration step:
14. pour adequate ddH2O or TE buffer into DNA tube & tap rapidly until
pellet dissolved. (if dissolved incompletely, heat 65C, 30min.)
protocol 2:
Cell Lysis Step:
1. 將全血離心3000 rpm，10 min.。
2. 加 RBC lysis buffer至玻璃試管2/3的高度，
吸取buffy coat，mix，靜置10 min.。
3. 離心2000 rpm，3 min.。
4. 倒掉上清液，(留下WBC)，倒扣衛生紙吸乾，
輕敲玻璃試管底部，使pellet 散開。
5. 加cell lysis buffer 600ul，vortex混合均勻(要完全)，
蓋上鋁箔紙，置入60℃烘箱 20 min.。
Protein Precipitation Step:
6. 加入10M NH4OAc 200ul，vortex混合均勻(要完全)(清澈)，
transfer到新的eppendorf。
7. 離心10000rpm，10min.。
DNA Precipitation Step:
8. 取上清液到新的eppendorf，加等量(800ul) isopropanol或加到9分滿。
9. inverse slowly直到白色絲狀物出現(太用力DNA會斷)
10. 離心12000rpm，10 min.。
11. 倒掉上清液，加入1ml 70% 酒精wash，
invert 使pellet懸浮。(刮eppendorf)
12. 離心12000 rpm，5 min.。
13. 倒掉上清液，倒扣eppendorf陰乾pellet。(放烘箱加快)
DNA Hydration Step:
14. 完全乾燥後視DNA量，加100~300ul ddH2O，
(DNA量少則加50ul)，加熱37~45℃溶解DNA，30min.。
15. -20℃儲存 / 取1ul DNA以2% agarose gel check。
