polydI-dC 是用来mimic viral dsRNA片段 一般用来作TLR3 activation的positive control 观察的基因表现 必定是TLR3 downstream molecules (ex. NF-kappaB) 回到这篇paper 这个实验大概是要问TLR3的"基因表现"是被哪一个pathway调控: 1) directly downstream of TLR3 pathway virus infection->TLR3 activation->increased TLR3 expression or 2) indirectly downstream of TLR3 pathway virus infection->???->increased TLR3 expresion 实验的设计就是比较 a) cell+ poly (dI-dC) -TLR3 related activation b) cell infected with virus (TLR3 and TLR3-independent) 结果 看到poly (dI-dC)这个TLR3 activation的positive control 并没有增加TLR3的基因表现 只有virus-infected cells 才会有TLR3表现增加的情形 所以结论：epithelial cells的TLR3基因表现 并不是经由TLR3 pathway调控 推测是由cytokine receptor等等调控 ※ 引述《[email protected] (minus3)》之铭言： : 请问poly(dI-dC)一般在观察基因表现实验中 所扮演的角色是什麽? : 原理又是什麽呢?? : 最近看PAPER看到的 我节录一些片段 : "TLR3 did not show induction by poly(dIC), whereas it showed : specific induction in infected cells." : "Controls included poly(dI-dC) and the C6/36 insect cell : supernatant used for production of D2V (mock infection)." : 虽然在第2个实验中 他有说是control可是为什麽可以做ccontrol不懂 : 而且感觉跟另一个实验使用目的不同 : 我翻了好几本书找不到 @@" : 不知道该怎麽找这个资料比较好 请各为大大解惑 : 感谢^^ --
