标题[方法] 请问有没有人做过peptide binding然後作Flow?

是这样的 目前是研究依一个secreted protein
觉得它应该会bind 某receptor
老师希望可以找具有此receptor的 cell population
所以 目前想 把这 protein去和cell binding
然後染此protein以及几个surface marker
上flow看一下这个ligand会bind哪种cell
请问有人做过类似实验吗
目前是 a. protein binding RT 1hr
b. 2.4G2 blocking on ice 15min
c. 一抗(染此protein) RT 1hr
d. 二抗 RT 30min then surfacr marker Abs 30min on ice
实验结果 减加protein binding没差异 都没有binding
有学长认为不要室温 receptor会endocytosis 所以bind不到
但 记得好像on ice receptor和ligand应该不会bind吧 ??
有先进有好建议吗
目前是想缩短染色时间 感觉染太久也对细胞不好 死亡率会提高
