这是英文的定义
The concept of the threshold cycle (Ct) allows for accurate
and reproducible quantification using fluorescence based
RT-PCR. Fluorescene values are recorded during every cycle
and represent the amount of product amplified to that point
in the amplification reaction. The more templates present at
the beginning of the reaction, the fewer number of cycles it
takes to reach a point in which the fluoresent signal is first
recorded as statistically significant above background, which
is the definition of the (Ct) values.
有看没有懂？根据我去Roche上课的记忆.....应该是曲线一次微分（即斜率）
的最大值.....因为软体会帮你算....所以很多人都只知道收data却不知原理
