标题[求救] IF染integrin beta-1 问题求救

各位强大的板友好~
实验室想要看材料上面的coating层对於
mesenchymal stem cell 贴附会造成甚麽样的影响，
因此有一部分的实验是利用免疫萤光染色观察
与focal adhesion 相关的protein 表现，
其中integrin β-1我试了许多条件
染完後在显微镜下观察都发现讯号非常的弱，几乎看不太到，
另外integrin的表现位置应该要在细胞膜上，
但我染出来是连整个cytosol都有看到，
不知道有没有大大染过相同的蛋白，
因此在这边想向大家求救，以下是我的实验条件:
1. Cell 培养於玻片上4小时候观察 (染色前有以光学显微镜确认细胞玻片上状况)
2. 4% paraformaldehyde (in PBS), RT, 15mins
3. 0.3% Triton x-100, RT, 10mins,
4. 5% goat serum, blocking for 1 hour, at RT
5. 1st Ab, 稀释比例-1:500, (4℃, 18hr; RT, 1.5hr and 37℃ 1hr 共三个条件)
6. 2nd Ab,稀释比例-1:100 RT, 1hr
1st Ab: Mouse monoclonal [12G10] to integrin beta 1(abcam, ab30394)
2nd Ab: Goat anti-mouse IgG (H&L)-FITC (CHEMICON, AP124F)

1F：推 jabari: 没染过 可是他在膜上 要不要试试不加triton?? 12/17 00:07
