标题[讨论] Gal4/UAS 系统中 UAS 的位置

我是做斑马鱼的
最近想做 Gal4/UAS 的 transgenic fish
原则上是采用 Tol2kit

以 three-insert multisite Gateway cloning 建构 plasmid
(insert 1: UASx10, 2: coding region 3:poly-A signal)
其实也不算难
但是老板想要更快，说不妨直接合成需要的序列丢进 destination vector
这样做更轻松 老板肯花钱我当然是爽
可是可是，问题来了：
标准的 three-insert gateway cloning 出来的结果，
insert 跟 insert 间当然会夹一些剩余的 recombinate site 之类的东西
（以我的目的来说就是 UASx10 跟 coding region 之间）
合成的 DNA 序列当然可以没有这些 construction 的残骸
但我却担心 UAS 跟 coding region 之间是否其实是需要一点距离的？
另外我的理解是一般的 promoter 已经内含 transcription start site (TSS)
但是 UAS 是 enhancer, 那麽 UAS 之後还需不需要外加 TSS 呢？
