标题[求救] Transient Assay 怪怪的

各位大大好
我最近在试transient assay
Plasmid定序都没问题
GFP 跟我的gene有 in frame
Promoter则是用ubiquitin promoter
可是试了用基因枪打洋葱表皮细胞
还有用叶片protoplast 作PEG transformation
隔天都看不太到GFP 的讯号
放到第二天才勉强可以看到微弱讯号
实验室学长姐说有可能是protein folding 太慢
想请问大家可能会是什麽原因造成的@@
如果是folding的问题又要怎麽解决呢？
