Note: Due to its low solubility in water, CDNB may cause the solution to become slightly cloudy. The solution should clear upon mixing. 3.Transfer the above solution into UV-transparent cuvettes which had been added 50 l GST enzyme (Note: the blank tube without enzyme). 4.Cover each cuvette with wax film and invert to mix (optional). 5.Place these cuvettes in a UV spectrophotometer blank at 340nm. Measure the absorbance of the sample cuvette at 340nm and simultaneously start a timer. 6.Record the absorbance readings at 340nm at one-minute intervals for 5 minutes. Note: The reaction rate is linear provided an A340 of approximately 0.8 is not exceeded during the five-minute time course. Initially, results may be plotted to verify that the reaction rate is linear over the time course.
